Jurkat cells were incubated and cultured in medium with (10 mM 2-DG (2-Deoxy-D-glucose)) or without 2-DG (Untreated control) for 60 min, washed and then incubated with 2-NBDG Glucose Uptake Assay Kit for 60 min, and flow cytometric detection of The glucose uptake of the cells was detected by flow cytometry. Incubation with 2-DG inhibited the glucose uptake of Jurkat cells.

Hela cells were incubated and cultured with (left) or without 10 mM 2-DG (right) for 60 min, washed and then incubated with 2-NBDG Glucose Uptake Assay Kit for 60 min, and glucose uptake was detected by fluorescence microscopy, and glucose uptake of Hela cells was inhibited by the addition of 2-DG incubation.

| Test Principle | 2-NBDG Reagent is a green fluorescently labeled deoxyglucose analog with the same transporters (GLUTs) and similar transport kinetics as D-glucose.It is allowed to be phosphorylated by hexokinase because of a modification at the C-2 position. When it enters the cell, it is phosphorylated by hexokinase at the C-6 position to produce 2-NBDG-6-phosphate (a fluorescent derivative). Thus the fluorescence intensity reflects the glucose uptake. |
